Establecimiento de una metodología para la inducción de regenerantes de arveja (Pisum sativum L.) variedad 'Santa Isabel'* Establishing a methodology for inducing the regeneration of pea (Pisum sativum L.) explants, 'Santa Isabel' variety
The effect of different 2,4-D, BAP and 2- ip concentrations on inducing the regeneration of peas from different types of explants was evaluated for use in genetic transformation as part of a project aimed at introducing resistance to Fusarium oxysporum f. sp. pisi which causes vascular wilt disease. Several types of explants were tested: cotyledons in abaxial and adaxial position, hypocotyls, epicotyls and zy- gotic embryos obtained from dry seeds and young seeds. These explants were evaluated in interaction with growth regulators in two types of light/cultiva- tion conditions, the first being a 16-hour light and 8- hour darkness photoperiod and the second consist- ing of total darkness. Three growth regulators were tested (2,4-D, BAP and 2-ip) in three combinations and different concentrations. Zygotic embryo regen- eration was obtained two light conditions, especially in the photoperiod and when applying BAP at 1 and 2.5 mg· L -1 concentrations.
