DETERMINACI"N DEL CONTENIDO DECIDO ASC"RBICO EN UCHUVA (Physalis peruviana L.), POR CROMATOGRAF˝A L˝QUIDA DE ALTA RESOLUCI"N (CLAR) ASCORBIC ACID DETERMINATION IN GOOSEBERRY (Physalis peruviana L.), FOR HIGH-PERFORMANCE LIQUID CHROMATOGRAPHY (HPLC)
The ascorbic acid (AA) it is an essential vitamin in the human diet, and their determination for sensitive and quick technical, it is important to evaluate their stability in different foods. At the moment the search of natural sources of AA, had great interest for the antioxidants characteristics of the vitamin; the gooseberry (Physalis peruviana L.), tropical fruit of the South American Andes, registers contents from 20 to 32 mg of AA for each 100 g of pulp, being this way, a promissory cultivation to include in the registrations of the sources common of this vitamin. The main objective of this study, was to find the optimize conditions for the extraction, identification and quantification by high-performance liquid chromatography (HPLC) of the present AA in the gooseberry (Physalis peruviana L.); for they were analyzed it different reported methodologies, for the extraction process. The phosphoric acid method, of they were recovery percentages between the 92,36 and 100,4 %; the most appropriate operation conditions for the quantification of AA for HPLC, were: Mobile phase: NaH2PO4 to 1 % pH = 2,7; Stationary phase: Hypersil C18 ODS 5μ m x 4,0 mm x 250 mm; Maximum absorption wavelength of: 265 nm and a Flow: 0,9 mL/min. The quantification methodology presented linealidad, precision, accuracy and sensibility at a level of trust of 95 %. The content of certain AA for the gooseberry was of 0,3320 mg (± 0,0262)/g sample, the standardization of the method of quantification of the AA for HPLC offers the opportunity to carry out later investigations about the stability of the vitamin in the gosseberry.
