Multiplicación y germinación de embriones somáticos en las variedades cubanas de soya Incasoy-1 e Incasoy-27

Abstract:

Soybean is genotype-dependent as it has been shown. This has lead to work on new protocols for plant regeneration. This research aimed to achieve growth and germination of somatic embryos in the Cuban varieties Incasoy-1 and Incasoy-27. Somatic embryos in globular stage (100 mg) were used as initial plant material. These were obtained after four weeks of culture in semisolid culture medium. Morphological characterization of somatic embryos was performed after 30 days. Then, the increase in fresh weight (mg) was determined. The percentage of germination was calculated in the germination phase and the number of somatic embryos that germinated on 100 mg was quantified. The growth and germination of somatic embryos in the Cuban varieties of soybean was achieved for the first time. Differences among them were observed regarding increasing of fresh mass during multiplication. Incasoy-1 and Incasoy-27 were higher than William-82. Over 15% of somatic embryos showed abnormal morphology in all varieties during germination. These were excluded from the count of somatic embryos in advanced stages of development since this is detrimental to the germination and plant regeneration. Incasoy-1 and Incasoy-27 were superior to Williams-82 taking into account percentage of germination and number of somatic embryos germinated.
