To investigate the effect of A561C (S128R) polymorphism in exon 4 of E-selectin gene(SELE) on blood pressure. Genotypes of 347 essential hypertension patients (EH group: male 163, female 184, age 46.08±12.49 years, BMI 26.13±3.14 kg/m2) and 157 healthy normotensives (NT group :male 93,female 222, age 42.21±14.67, BMI 25.66±3.19kg/ m2) were typed by polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP). Take blood pressure and exam BG, TG, TC, LDL, HDL, UA, BUN of each individuals. Three genotypes (AA,AC,CC) were found. The allele frequencies of C in hypertensive subjects were significantly higher than those in normotensive subjects(4.76% versus 2.22%, P=0.013).This coding variant, A561C (S128R), showed a strong association with essential hypertension . Risk of hypertension for allele C is 2.197 times of that for allele A(odds ratio, 2.197 [95% confidence interval, 1.164 to 4.144]. DBP and MAP were greater in individuals carrying the C allele(ρ = 0.024 for MAP and 0.049 for DBP), It seemed SBP a little higher in carriers of the C allele than in AA homozygotes, but was not significantly different. These results suggest association of A561C polymorphism in the E-selectin gene with blood pressure of essential hypertension patients. And it suggests that A561C polymorphism of SELE is a genetic susceptibility factor for essential hypertension.
